I don't use a handlebar bag, but here's what my backpack usually contains:

Sunscreen
Snacks
Water
U-lock and cable
A small amount of TP and some Purell (mostly as a charm to appease the gods of unplanned rest stops)
Pump
Spare front/rear lights
batteries
Tubes
Tire levers
Multi-tool
Chain-tool
Spoke wrench
Patch kit
Adjustable Wrench
15mm wrench
A couple of loose allen keys
Pliers
Lube

(And this is just for riding around town. I'd probably need to borrow Rich's extra-cycle if I ever decided to go touring.)

I have some first aid stuff in the front pocket along with a patch kit (the kit also has $6.50 so I have emergency tube money and enough to get home by bus as well as 2 5mm allen bolts in case one gets loose on the bike) and spare rechargeable batteries for the lights.
my back right pocket carries my alien II , and a small REI set of folding pliers (leathermanesque), my left back pocket carries my cell phone and a spare cell battery. the side pockets usually have a cliff bar and sometimes a pen and notepad.
The main part I switch things around in, most days you'll find lunch in there, maybe arm warmers or a jacket, if i'm going any distance it'll have a spare tube, then whatever shiney things catch my eye along my ride usually end up in there, currently there is only a sparkly cape in there.

I don't have a handlebar bag.

In my hiking-and-biking backpack I carry:
sunscreen
a fork, so I don't have to use disposable plastic ones
"unplanned rest-stop supplies", as JB puts it
an SD-memory case containing a few Naproxen and a few Benadryl. The former is good at controlling swelling when taken soon after an injury; the latter is first aid for allergic reactions. I figure that anything else first-aid related is either unlikely to prevent serious injury, or too heavy/bulky to carry around.

I don't use a handlebar bag either but when I am touring I keep a bunch of crap in my jersey pockets:
-Cortisone cream, you can apply it while on the move to both saddle sores and bug bites alike.
-Gels of some variety, Cliff or Hammer usually.
-Banana
-Cliff bars
-Snickers bars
-Chicken tenders
-Pocket knife for cutting open boils, or lemons I find to squeeze in to my water bottle.
-Fizzy tabs
-Toothbrush
-ipod in waterproof bag/case thingy
-Phone
-ID, insurance card, and credit card
-Sometimes a windbreaker
-Multi tool to make on the fly adjustments
-Small squeeze packs of chamois butter.
